Developing patient-tailored targeting of anti-inflammatory pathways in HFpEF

HFpEF, of Heart Failure with preserved Ejection Fraction, is een vorm van hartfalen waarbij het hart stijver is en zich niet goed vult. Het wordt ook wel diastolisch hartfalen genoemd. HFpEF behoort tot een grotere familie van cardiometabole ziekten (CMD). Deze ziekten ontstaan als gevolg van metabole ontregeling zoals die optreedt bij diabetes, obesitas of hyperlipidemie. Ze worden ook gekenmerkt door het feit dat ze worden veroorzaakt door een milde chronische ontstekingstoestand.
Hoewel HFpEF momenteel als één ziekte wordt gediagnosticeerd, zijn er verschillende clusters van aandoeningen die aanleiding kunnen geven tot diastolisch hartfalen. Het HFpEF-DIVA-consortium bestaat uit een klinische partner die zal werken aan het fenogroeperen van patiënten op basis van onbevooroordeelde clusteralgoritmen, en drie fundamentele wetenschappelijke groepen die elk een model van HFpEF zullen ontwikkelen om elke patiëntengroep te vertegenwoordigen. Alle fundamentele partners zullen dezelfde uitlezingen uitvoeren om te vergelijken hoe de verschillende diermodellen zich tot elkaar verhouden. Vervolgens zullen we de ziekten op verschillende manieren targeten op basis van het inflammatoire fenotype van elk model.
Verantwoordelijkheid
• Je zult ervaring opdoen met diermodellen van diastolisch hartfalen, evenals de beoordeling van het fenotype met behulp van echocardiografie, bloeddrukmetingen, bloedafname en inspanningstolerantietesten.
• Je voert histologische, beeldvormende en moleculair biologische technieken uit (Western blotting en/of ELISA's).
• Je bent verantwoordelijk voor de administratie en uitvoering van het project.
• Sterke vaardigheden op het gebied van moleculaire biologie en celcultuur zijn nuttig.
• Er wordt van je verwacht dat je het werk presenteert op conferenties, laboratoriumbijeenkomsten en afdelingsvergaderingen.
Dr. Elizabeth Jones en Dr. Stephane Heymans zijn op zoek naar een PhD-kandidaat werkzaam in het Departement Cardiovasculaire Wetenschappen van de KU Leuven. Je wordt ingeschreven in het PhD-programma van de KU Leuven en wordt mede begeleid door Drs. Jones en Heymans. Het project maakt deel uit van een groter Europees consortium (HFpEF-DIVA) en zal uitgebreide interacties met partners in Spanje en Frankrijk met zich meebrengen.Als promovendus word je in eerste instantie voor 12 maanden in dienst genomen. Dit zal bij bevredigende voortgang met nog eens drie jaar worden verlengd (voor een totaal van vier jaar).-
