PhD project: B cells and immunoglobulins in alcohol-associated liver disease

Er is een belangrijke rol van darmbacteriën bij het ontwikkelen van alcohol-gerelateerde leverziekte (ALD). Ten gevolge van darmschade door alcohol, lekken darmbacteriën en hun producten naar de bloedsomloop en de lever waar ze schade veroorzaken. Om ons te beschermen tegen binnendringende ziekteverwekkers, produceert ons lichaam verscheidene antilichamen (oftewel immunoglobulinen) die vreemd materiaal kunnen opruimen. Interessant is dat ALD-patiënten een verhoogde antilichaam-hoeveelheid in hun bloed vertonen, wat wijst op een veranderde immuunrespons op specifieke stoffen. Toch blijft hun exacte bijdrage aan ALD onduidelijk.
We veronderstellen dat verhoogde hoeveelheden van antilichamen in de bloedsomloop bij ALD patiënten een weerspiegeling is van veranderde immuniteit tegen darmmoleculen die leverbeschadiging veroorzaken. Daarom plannen we antilichamen, hun doelwitten en de cellen die ze produceren (B-cellen) in het bloed en de lever van ALD-patiënten te karakteriseren. Bovendien zullen we hun functionele rol tijdens ALD testen in geschikte muizenmodellen. Alles bij elkaar zullen deze studies nieuwe inzichten opleveren in de ziektemechanismen van ALD, met de identificatie van nieuwe aangrijpingspunten voor therapie.De afdeling Hepatologie aan de KU en UZ Leuven is een gerenommeerde eenheid binnen de dienst maag-, darm- en leverziekten. Deze eenheid staat bekend om zijn uitstekende patiëntenzorg en geavanceerde onderzoek naar lever-, galblaas- en pancreasziekten. Opgericht in 1963, geniet het laboratorium internationale faam en is het een van de langst bestaande levercentra ter wereld.De focus van het laboratorium ligt op een geïntegreerde aanpak die dagelijkse medische zorg combineert met translationeel onderzoek. Patiënten hebben hier toegang tot enkele van de meest vooruitstrevende geneesmiddelen en therapieën die momenteel beschikbaar zijn. Het team van specialisten is betrokken bij diverse klinische studies en wetenschappelijk onderzoek, wat bijdraagt aan de voortdurende verbetering van de behandeling van levergerelateerde aandoeningen. De functie binnen dit laboratorium zou zich richten op het bijdragen aan deze missie door middel van onderzoek, samenwerking met andere experts, en het delen van kennis op nationale en internationale platforms. Het is een kans om deel uit te maken van een toonaangevend team dat streeft naar innovatie en uitmuntendheid in hepatologie.Een volledig gefinancierde PhD positie voor vier jaar, mits gunstige evaluatie na 1 jaar.
